CAG Promoter Activity and Reprogramming

Writing in a recent STEM CELLS article, researchers led by Shangqin Guo (Yale University, New Haven, CT, USA) report a novel and simple methodology for the identification and enrichment of somatic cells correctly undergoing reprogramming into induced pluripotent stem cells. Hu et al. evaluated cell cycle, cell size, and transcriptome during reprogramming and linked the success of this process to the low activity of CAG, a widely used synthetic promoter. The simplicity of this approach may lead to widespread application in the reprogramming field.
